How do I adjust the sensitivity for optimum results in fluorescence or luminescence?

For the best fluorescence and luminescence results, make sure that you use the full dynamic range of the instrument. The easiest way to do this is to “scale to high well” if you have a known positive control or “high standard” on your plate. Most of our readers have a maximum of 99,999 counts, so a good target for the high well is 80,000. This will give a little room if there are other wells with more signal. If you find that many wells are overranging, you can reduce the target value. After performing the read this way, look at the signal in your blank wells. Good blank readings should be less than 500 RLU in luminescence and less than 10,000 RFU in fluorescence. Adjust your sensitivity down to lower the blank readings.
