Do I have to use a blank on the plate when quantitating DNA at 260 nm?
A blank is required on the plate when quantitating DNA at 260 nm. The blank well should contain the same buffer, usually deionized water, contained in the sample wells.
A blank is not required if using a blank plate which is subtracted from the assay plate. However, a blank well or two on the assay plate is usually sufficient.
